DNA extraction from sediments

For the extraction of genomic DNA from environmental samples, we use a protocol modified after Zhou et al. (1996).

Reagents and Solutions:

DNA-extraction buffer:
100 mM Tris-HCl (pH 8.0)
100 mM EDTA (pH 8.0)
100 mM Na-phosphate (pH 8.0)
1.5 mM NaCl
1% CTAB (Hexadecyltrimethylammonium bromide)


10 mg/ml proteinase K

Chloroform-isoamyl alcohol (24:1 v/v)

20% SDS

Isopropanol

80% ethanol
